Anti-SARS-CoV-2 IgG from severely ill COVID-19 patients promotes macrophage hyper-inflammatory responses

By: biorxiv.org

Please read about this important findings in a Dutch study report:

- anti-Spike IgG from serum of severely ill COVID- 19 patients induces a hyper-inflammatory response by human macrophages.

- differences in the glycosylation pattern of the Fc region of these antibodies, which are responsible for the inflammatory responses observed in vitro.

- findings recapitulate the exacerbated immune response observed in severely ill patients, which could play a role in the pathology of the disease.

- Note  of caution about the therapeutic use of convalescent plasma from severely ill patients.

- identified biomarkers that could be used to monitor these “toxic” antibodies during disease, and triage plasma samples from these patients for therapeutic use.

- Syk inhibitors, some FDA and EMA approved, block the hyperinflammatory response, and they could potentially be repurposed for treatment of severe COVID-19.
